The guys see the hotel:
Dean: Dude, this is sweet! I never get to work jobs like this!
Sam: Like what?
Dean: Old school haunted houses. You know – fog, secret passageways, sissy British accents… we might even run into Fred and Daphne inside. [He gets a lustful look in his eyes.] Mmmm. Daphne. Love her.
And we love your one-track mind, Dean.
Dean and Sam discuss their greeting at the reception desk:
Dean: Of course, the most troubling question is, why do these people assume we're gay.
Sam: Well you are kind of butch – they probably think you're overcompensating.
Or, it could be that two ridiculously good-looking guys checking into a hotel together tends to mean nookie. We're just saying….
Dean and Sam try to get into Susan's office:
Dean: Are those antique dolls? Cause this one, this one here, he's got a major doll collection back home, huh?
Sam [hesitating]: Big time.
Dean: Big time! You think he could come in, well, we could come in and take a look?
Susan: I don't know…
Dean: Please? I mean, he loves them. He's not going to tell you this, but he's always dressing them up in these little tiny outfits – you'd make his day. She would Huh? Huh?
Sam [woodenly.] It's true.
[Susan ushers them in]
Dean: Wow! This is a lot of dolls. And, they're nice, you know? Not super creepy at all!
Heh. The expressions on Sam and Dean's faces during this whole exchange were fantastic. Dean was nearly delirious with glee at being able to cause Sam such obvious discomfort, while Sam looks like he wants to burst out with "Dude! I do not play with dolls!" But for Susan's benefit, he needs to play along. We laughed so hard we had to rewind and watch it about five times before moving on to the next scene.
Dean enters the room to find Sam slouching in the dark. Dean asks Sam a question.
Sam: You're bossy.
Dean: What?
Sam: You're bossy… and short.
Drunken Sam rocks! Next time, can we see drunken karaoke? We'll be your best friend!
The guys find Grandma Rose in the attic. She's immobile. Dean thinks maybe she's faking the stroke.
Sam: What are you going to do, poke her with a stick?
[Dean makes a face like he's thinking about it.]
Sam: Dude, you're not going to poke her with a stick!
Because you know that if there had been a stick handy, Dean would have started poking.
